顺铂是一种较好的抗肿瘤药,但其肾和神经毒性限制了它的用途.一般抗肿瘤药包裹在脂质体中能降低毒性,并保持其抗肿瘤作用.本文试图提高顺铂的包裹率,增加脂质体的抗渗漏能力,同时作体外抑制肿瘤细胞生长试验.多层脂质体用经典薄膜法制备,磷脂双分子层组成为PC/PS/Chol(10:1:4)或DPPC/DPPG/Cho(10:1:10).脂质体用动态光散射法测定平均粒径和分散系数,微电泳测定ζ电位,铂含量用原子吸收光谱测定.体外用腹水型胃鳞状肿瘤细胞株5D04,以〔~8H〕胸腺嘧啶渗入法测定顺铂对肿瘤细胞的抑制生长百分率(GI%).
Cisplatin is a good anti-tumor drug, but its renal and neurotoxicity limits its use.Generally anticancer drug encapsulated in liposomes can reduce the toxicity and maintain its anti-tumor effect.This paper attempts to improve cisplatin , And increase the anti-leakage ability of the liposomes, meanwhile, it can inhibit the growth of tumor cells in vitro.Multi-layer liposomes were prepared by the classical membrane method.The composition of the phospholipid bilayer was PC / PS / Chol (10: 1: 4) or DPPC / DPPG / Cho (10: 1: 10). The average particle diameter and dispersion coefficient of liposomes were determined by the dynamic light scattering method, the zeta potential was determined by microelectrophoresis and the content of platinum was determined by atomic absorption spectrometry. Gastric squamous cell carcinoma cell line 5D04 was treated with [~ 8H] thymidine infiltration method to determine the percentage inhibition (GI%) of cisplatin on tumor cells.
