金丝桃苷对Aβ_(1-42)介导的血脑屏障体外模型损伤的影响

目的:探讨金丝桃苷(hyperoside,Hyp)对淀粉样蛋白(Aβ_(1-42))介导的血脑屏障(blood-brain barrier,BBB)体外模型损伤的影响。方法:本研究以Hyp干预纤维状Aβ_(1-42)介导的b End.3细胞与原代星形胶质细胞(astrocytes,As)BBB体外共培养模型的损伤,通过检测各培养组荧光素钠的通透性,比较各组BBB渗透性的变化,评价Hyp对Aβ_(1-42)介导的BBB的通透性的影响;采用噻唑蓝(MTT)比色法检测Hyp对Aβ_(1-42)介导的b End.3细胞损伤的细胞活力影响;再以蛋白免疫印迹法(Western blot)检测相关凋亡蛋白,初步探索其保护机制。结果:与模型组比较,Hyp低、中、高浓度组均能降低BBB的通透性(P<0.05,P<0.01);Hyp中、高浓度组能显著提高b End.3细胞活性(P<0.05);Western blot结果显示,经Hyp预处理的组别中,Bax/Bcl-2,cleaved半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)/Caspase-3的及细胞色素-C(Cyt-C)的表达量均明显下降,且差异均具有统计学意义(P<0.05)。结论:Hyp通过保护脑微血管内皮细胞抑制纤维状Aβ_(1-42)诱导的BBB通透性增加。 Objective: To investigate the effect of hyperoside (Hyp) on the injury of amyloid protein (Aβ 1-42) -mediated blood-brain barrier (BBB) ​​in vitro. Results: Compared with the model group, Hyp low, middle and high concentration groups could reduce the permeability of BBB (P <0.05, P <0.01); Hyp and high concentration groups could significantly increase the activity of b End.3 cells (P <0.05). Western blot results showed that in the groups pretreated with Hyp, the expressions of Bax / Bcl-2, cleaved Caspase-3 / Caspase-3 and cytochrome- C (Cyt-C) expression levels were significantly decreased, and the differences were statistically significant (P <0.05). Conclusion: Hyp suppresses the increase of BBB permeability induced by fibrillar Aβ 1-42 in brain microvascular endothelial cells.
