目的 研究多药耐药相关蛋白 (multidrugresistanceassociated protein ,MRP)基因、肺耐药蛋白 (lungresistanceprotein,LRP)基因在肝细胞癌 (hepatocelluarcarcinoma,HCC)中的表达水平与HCC病人化疗疗效及甲胎蛋白 (a fetoprotein ,AFP)的关系。方法 采用S P免疫组织化学方法和原位PCR技术对 54例术前未化疗的未能切除的HCC ,2 4例肝癌旁及 12例肝炎后肝硬化存档石蜡包埋切片组织中的MRP、LRP基因编码的MRP、mRNAMRP和LRP、mRNALRP的表达进行检测。术后所有病人常规化疗 ,分析MRP、LRP基因表达与HCC化疗疗效关系。并对其术后 2周AFP阳性的 38例HCC病人化疗 ,分析MRP、LRP基因表达与AFP变化的关系。结果 MRP、mRNAMRP和LRP、mRNALRP在三种组织中的阳性表达率分别为 6 1 1% ,2 5 0 % ,16 7% ;77 8% ,37 5% ,4 1 7%和 6 4 8% ,33 3% ,2 5 0 % ;75 9% ,33 3% ,2 5 0 %。HCC组织中MRP、LRP基因表达显著高于其他组织中的基因表达 (P <0 0 5,0 0 5)。HCC组织中MRP、LRP基因表达与LRP基因表达呈正相关 (r =0 2 87,P <0 0 5;r =0 32 4 ,P<0 0 5)。MRP、LRP基因表达阴性组HCC病人化疗有效率显著高于阳性组病人 (P <0 0 5,0 0 5)。MRP、LRP基因表达阴性组病人 ,AFP有效率高于阳性组 (P <0 0 5)。结论?
Objective To investigate the expression of multidrug resistance associated protein (MRP) gene, lung resistance protein (LRP) gene in hepatocellular carcinoma (HCC) fetoprotein, AFP). Methods The expression of MRP and LRP genes in 54 cases of unresectable HCC, 24 cases of HCC and 12 cases of posthepatitic cirrhosis with paraffin embedded sections were detected by SP immunohistochemistry and in situ PCR MRP, mRNA MRP and LRP, mRNALRP expression were detected. All patients after conventional chemotherapy, analysis of MRP, LRP gene expression and the relationship between the efficacy of HCC chemotherapy. Chemotherapy was performed on 38 AFP positive HCC patients after 2 weeks of operation. The relationship between MRP, LRP gene expression and AFP was analyzed. Results The positive rates of MRP, mRNA MRP, LRP and mRNALRP in the three tissues were 61.1%, 25.0%, 16.7%, 77.8%, 37.5%, 41.7% and 64.8% , 33 3%, 25 0%; 75 9%, 33 3%, 25 0%. The mRNA expression of MRP and LRP in HCC tissues was significantly higher than that in other tissues (P <0.05 05,05). There was a positive correlation between MRP and LRP gene expression and LRP gene expression in HCC (r = 0 2 87, P <0 05; r = 0 32 4, P 0 05). The effective rate of chemotherapy for HCC patients with negative MRP and LRP gene expression was significantly higher than that of positive patients (P <0.05,0.05). The effective rate of AFP in MRP and LRP gene negative patients was higher than that in positive patients (P <0.05). in conclusion?
