目的观察芪参益气滴丸对Ameroid缩窄环致心功能不全小型猪心功能及心室重构的影响。方法巴马小型猪22只,采用左冠状动脉前降支放置缩窄环的方式制备小型猪慢性心肌缺血心功能不全模型,随机分为模型组、地高辛组、芪参益气滴丸组,每组6只,另制备假手术动物4只。于术后第4周开始口服给药,地高辛组每日1次0.012 5 mg/kg,芪参益气滴丸组每日3次,每次0.025 g/kg,持续8周。于给药前和给药后2、4、8周末超声心动检测各组动物心功能,于给药后第8周末取材进行心肌组织病理观察。结果超声心动结果显示,给药前,模型动物左室射血分数(EF)显著下降(P<0.01);给药后2周,地高辛组动物左室舒张末容积(LVDV)、左室收缩末容积(LVSV)、EF显著改善(P<0.05);给药后4周芪参益气滴丸组动物LVDV、LVSV、EF显著改善(P<0.05或P<0.01)。HE染色显示,模型动物左心室心肌出现细胞结构异常和大量炎细胞浸润,芪参益气滴丸组心肌细胞排列紊乱程度减轻,炎细胞浸润程度轻于模型组。Masson染色显示,模型动物心肌胶原呈现弥漫性分布,芪参益气滴丸组胶原呈树突状分布,胶原含量少于模型组。结论芪参益气滴丸能够显著改善Ameroid缩窄环致心肌缺血心功能不全小型猪心功能并抑制心室重构。
Objective To observe the effect of Qishen Yiqi drop pill on heart function and ventricular remodeling in Ameroid narrowing ring induced heart dysfunction miniature pigs. Methods Twenty-two Bama miniature pigs were randomly divided into model group, digoxin group, Qishen Yiqi Dripping Pill Group, 6 in each group, and 4 sham-operated animals were also prepared. Oral administration began at 4 weeks after operation. The digoxin group received 0.012 5 mg / kg once daily and the Qishen Yiqi Drops group took 0.025 g / kg three times a day for 8 weeks. The heart function of each group of animals was detected by echocardiography before administration and at 2, 4, and 8 weeks after administration. The pathological changes of myocardium were observed at the end of 8th week after administration. Results Echocardiography showed that left ventricular ejection fraction (EF) decreased significantly in model animals before administration (P <0.01). After 2 weeks of administration, left ventricular end-diastolic volume (LVDV) (P <0.05). After 4 weeks of administration, the LVDV, LVSV and EF of Qishen Yiqi pills group were significantly improved (P <0.05 or P <0.01). HE staining showed that myocardial cell structure abnormalities and a large number of infiltration of inflammatory cells in the model animal left ventricular myocardial cells Qishen Yiqi pill group to reduce the degree of disorganization of myocardial cells, inflammatory cell infiltration was lighter than the model group. Masson staining showed diffuse distribution of myocardial collagen in the model animals, and the dendritic distribution of collagen in the Qishenyiqi dropping pills group was less than that in the model group. Conclusion Qishenyiqi dripping pills can significantly improve the heart function of Ameroid narrowing ring-induced myocardial ischemia heart failure and inhibit ventricular remodeling.
