目的:建立天钩降压胶囊内容物正丁醇萃取液的高效液相指纹图谱的检测方法。方法:采用高效液相色谱法,色谱柱TSKgel ODS-100V色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.1%磷酸水溶液,梯度洗脱,柱温30℃,流速1 m L·min-1,检测波长230 nm。结果:建立了天钩降压胶囊内容物正丁醇萃取液指纹图谱共有模式,并分别指认其中的4个色谱峰。通过与对照品的保留时间和紫外光谱图比较,正丁醇萃取液指纹图谱中6,8,9,14号峰的归属分别为:天麻素,松脂醇二葡萄糖苷,黄芩苷,紫丁香苷。10批天麻枣仁安神胶囊正丁醇萃取液的HPLC指纹图谱相似度均大于0.88。结论:该方法灵敏度高、稳定性和准确性良好,为天钩降压胶囊质量标准建立提供了可靠的科学依据。
Objective: To establish a HPLC method for the determination of n-butanol extract in Tian hookjiangya capsule. Methods: High performance liquid chromatography (HPLC) was used on a TSKgel ODS-100V column (4.6 mm × 250 mm, 5 μm) with a mobile phase of acetonitrile-0.1% phosphoric acid and gradient elution at 30 ℃ with a flow rate of 1 mL · min-1, detection wavelength 230 nm. Results: The fingerprint of n-butanol extract of Tian hookjiangya capsule was established and the four peaks were identified. Compared with the retention time and UV spectrum of the reference substance, the peaks assigned to peaks 6, 8, 9 and 14 in the n-butanol extract fingerprint were as follows: gastrodin, pinoresinol diglucoside, baicalin, . Ten batches of Tianmazaoan Anshen capsule n-butanol extract HPLC fingerprint similarity greater than 0.88. Conclusion: The method has high sensitivity, good stability and accuracy, and provides a reliable scientific basis for establishing the quality standard of Tian hook hypotensive capsules.
