目的 :研究人乳腺癌中p16基因mRNA及其编码蛋白的表达水平与肿瘤发生和预后的相关性。方法 :采用免疫组化和原位杂交方法分别对 11例乳腺良性肿瘤和 5 9例乳腺癌中的p16基因及其编码蛋白进行检测。结果 :p16基因蛋白表达水平与乳腺肿瘤的良、恶性显著相关 (P <0 0 5 ) ;p16基因mRNA及其蛋白表达水平与乳腺癌的腋窝淋巴结转移呈显著负相关 (P <0 0 5 ) ,而与乳腺癌的术后生存期呈显著正相关 (P <0 0 5 ) ;且免疫组化和原位杂交两种检测方法的结果呈显著关联 (P <0 0 1)。结论 :p16基因蛋白低表达与乳腺癌的发生有关 ;p16基因mRNA及其蛋白表达水平均可作为估价乳腺癌预后的重要指标
Objective: To study the correlation between the expression of p16 mRNA and its protein in human breast cancer and tumorigenesis and prognosis. Methods: Immunohistochemistry and in situ hybridization were used to detect p16 gene and its encoded protein in 11 cases of benign breast tumors and 59 cases of breast cancer. Results: The expression of p16 protein was significantly correlated with benign and malignant breast tumors (P <0.05). There was a significant negative correlation between p16 mRNA and protein expression and axillary lymph node metastasis in breast cancer (P <0.05) , But significantly correlated with the postoperative survival of breast cancer (P <0.05). The results of immunohistochemistry and in situ hybridization showed significant correlation (P <0.01). Conclusion: The low expression of p16 protein is associated with the occurrence of breast cancer. The expression of p16 gene mRNA and its protein may be used as an important index to evaluate the prognosis of breast cancer
