目的研究维生素D(VD)对大鼠单纯性肥胖的预防作用。方法将50只清洁级2月龄Wistar雄性大鼠随机分成5组,分别为对照组、肥胖组和低(0.6μg/ml)、中(1.2μg/ml)、高剂量(2.4μg/ml)VD干预组,每组10只。采用经口灌胃方式给予VD,灌胃容量为5 ml/kg,每天1次,连续灌胃9周。对照组饲喂普通饲料,其余组饲喂高脂饲料。灌胃结束后,肥胖组大鼠达到单纯性肥胖标准;测定大鼠体重、体长、附睾脂肪垫重量,并计算Lee’s指数和脂肪指数,采用高效液相色谱法测定血清VD含量。结果与对照组比较,肥胖组大鼠体重、Lee’s指数、附睾脂肪垫重量及脂肪系数均较高,血清VD含量较低,差异有统计学意义(P<0.05)。随着VD干预剂量的升高,大鼠的体重、Lee’s指数、附睾脂肪垫重量及脂肪系数呈下降趋势,血清VD含量均呈上升趋势。大鼠血清VD含量与Lee’s指数和脂肪指数均呈负相关(P<0.01)。结论 VD能够预防和控制大鼠单纯性肥胖。
Objective To study the preventive effect of vitamin D (VD) on simple obesity in rats. Methods Fifty clean Wistar male rats aged 2 months were randomly divided into 5 groups: control group, obesity group and low (0.6μg / ml), medium (1.2μg / ml) and high dose VD intervention group, 10 rats in each group. VD was given by oral gavage. The gavage volume was 5 ml / kg once daily for 9 weeks. The control group was fed normal feed, and the rest fed high fat diet. After the end of gavage, obese rats reached the standard of simple obesity. Body weight, body length and weight of epididymal fat pad were measured. Lee’s index and fat index were calculated. Serum VD levels were determined by HPLC. Results Compared with the control group, body weight, Lee’s index, fat pad weight and fat coefficient of epididymis in obese rats were higher than those in control rats, and serum VD levels were lower (P <0.05). With the increase of VD intervention dose, body weight, Lee’s index, fat pad weight and fat coefficient of epididymal were decreased, while serum VD content showed an upward trend. Serum VD levels in rats were negatively correlated with Lee’s index and fat index (P <0.01). Conclusion VD can prevent and control simple obesity in rats.
