目的 研究1 d中不同时间点过量锰的暴露对雄性大鼠血清中总胆汁酸(TBA)活性的影响.方法 健康成年雄性SD大鼠96只,在严格控制光照/黑暗(12 h/12 h)条件下适应性喂养7 d后,分别于1 d中的授时时间点2(ZT2)(9:00)、授时时间点8(ZT8)(15:00)、授时时间点14(ZT14)(21:00)与授时时间点20(ZT20)(3:00)进行腹腔注射剂量为30 mg/kg的MnCl2,对照组等剂量注射蒸馏水,1次/d,连续21 d.第22天,处死动物,获取动物血清,检测其TBA含量.结果 过量锰暴露可引起雄性大鼠TBA含量升高,其中在ZT2、ZT8与ZT20升高的最明显,分别升高0.79、5.70与6.23倍.余弦分析显示,锰暴露组的TBA日均值上升,振幅增加,峰值时间提前.结论 过量锰暴露均可引起血清中TBA的改变,其中在ZT20授时时间点改变最严重,其次为ZT8,第3为ZT2;对TBA改变最弱的为ZT14授时时间点.“,”Objective To study the chronotoxicity of manganese(Mn)on total bile acids(TBA)in male rats. Methods Ninety-six healthy SPF SD rats in 12 h:12 h light-dark photoperiod,which were divided into four Mn groups and four control groups. Mn groups were administered by intraperitoneal injection(i.p.)of Mn (30 mg/kg MnCl2 ·4H 2 O)and the control groups were administered with equal volume of water,respectively at corresponding zeitgeber time(ZT2,ZT8,ZT14,ZT20)for 21 days. The total bile acids(TBA)levels were measured by ELISA method. Results Compared with the control group,Mn exposure induced ChE level increased at ZT2,ZT8 and ZT20, increased 0.79times、 5.70times and 6.23 times respectively. Cosinor analysis showed that the mean value of TBA in the Mn exposure groups ascended,amplitude increased,the peak time was ahead of time. Conclusions Excessive manganese exposure may cause the change of TBA. Different exposure time would induce different result. Exposed to Mn at ZT20,the TBA change was most serious,secondly was at ZT8, thirdly was at ZT2.And, exposed at ZT14, the TBA change was the weakest.
