液相色谱-质谱联用(LC-MSn)的高灵敏度和高专属性在药物研究特别是药物代谢研究中发挥着重要作用。但是随着高活性低剂量药物和特殊结构类型化合物的出现,液质联用的应用也受到限制。通过衍生化改变药物的结构,从而改变其理化性质,在液质联用检测中可以提高离子化效率,降低基质抑制和减少无机盐及内源性杂质的干扰。本文简要综述了衍生化和液质联用技术应用于生物体液中药物定量检测灵敏度提高方面的最新进展。
The high sensitivity and high specificity of liquid chromatography-mass spectrometry (LC-MSn) play an important role in drug research, especially in drug metabolism research. However, with the emergence of high activity and low dose of drugs and special structure type compounds, LC / MS applications are also limited. By derivatization, the structure of the drug can be changed to change the physicochemical properties of the drug, which can improve the ionization efficiency and reduce the matrix inhibition and reduce the interference of inorganic salts and endogenous impurities in the liquid chromatography-mass spectrometry. This review summarizes the recent advances in the application of derivatization and LC / MS techniques in the quantitative detection of drugs in biological fluids.
