目的 :探讨三氟拉嗪 (trifluoperazine,TFP)对视网膜色素上皮 (retinal pigment epithelium,RPE)细胞生长、增殖以及细胞周期的影响 ,为 TFP治疗增殖性玻璃体视网膜病变 (proliferative vitreoretinopathy,PVR)提供理论依据。方法 :通过细胞生长曲线的测定 ,观察不同浓度 (5、 10、 15、 2 0 μmol/ L) TFP对 RPE细胞生长的影响 ;通过 MTT比色法测定不同浓度 (5、 10、15、 2 0 μmol/ L) TFP作用 RPE细胞 3天时的吸光值 A,并计算抑制率 ;通过流式细胞分析法检测 10 μmol/ L TFP作用 72小时时 ,RPE细胞周期的改变。结果 :给药三天后 ,各浓度 (5、 10、 15、 2 0 μmol/ L) TFP对 RPE细胞生长均具有明显抑制作用 ,且具有时间剂量效应。RPE细胞经 10 μmol/ L TFP处理 72 h后 ,经细胞荧光流速分析 ,结果显示 TFP使 G1 期细胞增加了 2 7.0 2 % ,S期细胞下降了 9.0 7% ,G2 M期细胞下降了 45 %。结论 :TFP可抑制 RPE细胞生长 ,且呈时间剂量效应。TFP通过对 RPE细胞周期各时相群体的影响 ,使 G1 期向 S期 ,进而向 G2 / M期的过渡被抑制 ,从而抑制 RPE细胞的增殖
Objective: To investigate the effect of trifluoperazine (TFP) on the growth, proliferation and cell cycle of retinal pigment epithelium (RPE) cells and to provide a theoretical basis for TFP treatment of proliferative vitreoretinopathy (PVR) . Methods: The effects of TFP (5, 10, 15, 20 μmol / L) on the growth of RPE cells were observed by measuring the growth curve of cells. MTT assay was used to determine the effect of different concentrations of TFP on the growth of RPE cells (5, 10, 15, μmol / L), the absorbance A of RPE cells at 3 days was calculated and the inhibition rate was calculated. The cell cycle changes of RPE cells were detected by flow cytometry at 72 hours after treated with 10 μmol / L TFP. Results: After three days of administration, TFP at various concentrations (5, 10, 15, 20 μmol / L) significantly inhibited the growth of RPE cells and had a time-dose effect. Flow cytometry analysis of RPE cells treated with 10 μmol / L TFP for 72 h showed that TFP increased cells in G1 phase by 7.07%, cells in S phase decreased by 9.0 7% and cells in G2 M phase decreased by 45% . Conclusion: TFP can inhibit the growth of RPE cells in a time-dose-dependent manner. TFP can inhibit the transition of G1 phase to S phase and then to G2 / M phase through the influence on each phase population of RPE cell cycle, and thus inhibit the proliferation of RPE cells
