为研究以2型登革病毒NS3基因构建的核酸疫苗能否诱导体液免疫,用表达2型登革病毒NS3的重组真核表达质粒PSV·NS3直接接种Balb/c小鼠。结果发现在接种后2周即可检测到IgM抗体,该抗体在加强接种后滴度显著升高,6周达最高水平至少维持2个月;IgG抗体在接种后4周可检测到,8周达最高水平至少维持2个月。Westernblot证实了接种小鼠血清中有NS3特异性的IgG抗体。这些结果表明我们接种的PSV·NS3能诱导Balb/c小鼠的体液免疫。
To investigate whether humoral immunity can be induced by nucleic acid vaccine constructed with Dengue virus type 2 dengue virus, Balb / c mice were directly inoculated with recombinant eukaryotic expression plasmid PSV-NS3 expressing Dengue virus type 2. As a result, it was found that IgM antibody was detected two weeks after inoculation. The antibody showed a marked increase in titers after boosting, reaching a maximum level of at least 6 months at 6 weeks. IgG antibodies were detected 4 weeks after inoculation and 8 weeks The highest level of at least 2 months. Westernblot confirmed the NS3-specific IgG antibodies in sera of mice vaccinated. These results indicate that we vaccinated PSV · NS3 can induce humoral immunity in Balb / c mice.
