目的 :TTNPB酸乙酯及其代谢物TTNPB同属维甲酸类药物 ,是治疗银屑病的有效药物。为了测定TTNPB酸乙酯及代谢物TTNPB在体内的血药浓度 ,特建立了反相高效液相色谱同时测定TTNPB酸乙酯和TTNPB的分析方法。方法 :反相高效液相色谱法 ,C18柱 ,流动相为甲醇 - 1%冰醋酸 ( 80∶2 0 ) ,流速为 1mL·min-1,柱温 3 5℃ ,检测波长 3 0 5nm。结果 :TTNPB酸乙酯在 5 0ng~ 5 μg范围内质量与峰面积呈线性 ,相关系数r =0 9992 ,检测下限 2 0ng ;TTNPB在 3 0ng~ 5 μg内质量与峰面积呈线性 ,相关系数r =0 9994 ,检测下限为 5ng。在血浆中TTNPB酸乙酯和TTNPB的回收率大于 90 % ,相对标准偏差小于 5 %。 结论 :该方法简便 ,稳定 ,可用于TTNPB酸乙酯及代谢物TTNPB的同时测定和药代动力学研究
Objective: TTNPB acid ethyl ester and its metabolites TTNPB belong to the same retinoid drugs, is an effective drug for the treatment of psoriasis. In order to determine the plasma concentration of TTNPB and its metabolite TTNPB in vivo, an analytical method was established for simultaneous determination of TTNPB and TTNPB by RP-HPLC. Method: RP-HPLC was performed on a C18 column with a mobile phase of methanol-1% glacial acetic acid (80:20) at a flow rate of 1 mL · min-1. The column temperature was 35 ℃ and the detection wavelength was 350 nm. Results: The mass and peak areas of TTNPB acid ethyl ester ranged from 50 ng to 5 μg with a correlation coefficient of r = 0 9992 and a detection limit of 20 ng. The linear relationship was found between the mass and peak area of TTNPB between 30 ng and 5 μg, r = 0 9994, the detection limit of 5ng. The recovery of TTNPB ethyl acetate and TTNPB in plasma was greater than 90% and the relative standard deviation was less than 5%. Conclusion: The method is simple and stable and can be used for the simultaneous determination and pharmacokinetics of TTNPB and its metabolite TTNPB
