以美国白蜡饱满种子和幼嫩茎段为外植体,研究了0.1%升汞不同消毒时间、不同类型增殖培养基和所添加激素种类、浓度对美国白蜡组培快繁效果的影响。实验结果表明:以饱满种子为外植体,0.1%升汞消毒10min,消毒无菌率达96.7%;最佳增殖培养基为MS+CP-PU0.15mg/L+NAA0.2mg/L,平均增殖系数达6.87;在1/2WPM+IBA1.0mg/L的生根培养基中,平均生根率达88.9%;生根苗驯化后移栽到装有基质细河沙?珍珠岩=2?1的育苗穴盘中,成活率在90%以上。
The effects of 0.1% mercuric chloride at different disinfection time, different types of proliferation medium and added hormones and concentration on the fast propagation of tissue culture of American white wax were studied.
