目的探讨在4种宫颈癌细胞系中特异表达的水通道蛋白(aquaporins,AQPs)亚型及意义。方法通过水通透性实验比较4种人宫颈癌细胞系SiHa、HeLa、Caski、ME-180的水通透性差异,从中选择水通透性低的细胞系作为实验用细胞系,应用荧光定量PCR、免疫荧光检测细胞系中特异表达的AQPs亚型。结果①SiHa中100 mOsm与300 mOsm、200 mOsm与300 mOsm组间水通透性差异无统计学意义(P>0.05),400mOsm与300 mOsm、500 mOsm与300 mOsm组间水通透性差异有统计学意义(P<0.05);Caski中100 mOsm与300 mOsm、200 mOsm与300 mOsm、500 mOsm与300 mOsm组间水通透性差异有统计学意义(P<0.05),400mOsm与300mOsm组间水通透性差异无统计学意义(P>0.05);HeLa中100 mOsm与300 mOsm、400 mOsm与300 mOsm组间水通透性差异有统计学意义(P<0.05),200 mOsm与300 mOsm、500 mOsm与300 mOsm组间水通透性差异无统计学意义(P>0.05);ME-180中各组间水通透性差异均有统计学意义(P<0.05);②SiHa中表达的特异AQPs亚型有AQP0、AQP1、AQP2、AQP3、AQP4、AQP5和AQP8。结论 SiHa细胞水通透性最低,在SiHa细胞中表达的AQP亚型可能与SiHa细胞的迁移及转移有关。
Objective To investigate the subtypes of aquaporins (AQPs) specifically expressed in four kinds of cervical cancer cell lines and their significance. Methods The water permeability of four human cervical cancer cell lines SiHa, HeLa, Caski and ME-180 were compared by water permeability test. The cell lines with low water permeability were selected as experimental cell lines, and the fluorescence quantitative PCR, immunofluorescence detection of cell lines specifically expressed AQPs subtypes. Results ① There was no significant difference in water permeability between 100 mOsm and 300 mOsm, 200 mOsm and 300 mOsm in SiHa (P> 0.05). There was statistical difference in water permeability between 400 mOsm and 300 mOsm, 500 mOsm and 300 mOsm (P <0.05). There was significant difference in water permeability between 100 mOsm and 300 mOsm, 200 mOsm and 300 mOsm, 500 mOsm and 300 mOsm in Caski (P <0.05), between 400 mOsm and 300 mOsm (P> 0.05). There was a significant difference in water permeability between 100 mOsm and 300 mOsm, 400 mOsm and 300 mOsm in HeLa (P <0.05), 200 mOsm and 300 mOsm, There was no significant difference in water permeability between 500 mOsm and 300 mOsm groups (P> 0.05). The differences of water permeability among ME-180 groups were statistically significant (P <0.05). ② The specific expression of SiHa AQPs are AQP0, AQP1, AQP2, AQP3, AQP4, AQP5 and AQP8. Conclusion SiHa cells have the lowest water permeability. The AQP subtypes expressed in SiHa cells may be related to the migration and metastasis of SiHa cells.
