目的探讨hMLH1基因在新疆维吾尔族食管癌组织中的表达及其在食管癌发生、发展中的作用。方法采用逆转录聚合酶链反应技术检测hMLH1基因在20例食管癌癌组织及其相应癌旁组织中的表达情况,并与临床病理资料结合分析其意义。结果 hMLH1在食管癌组织中表达阳性率为55%(11/20),癌旁组织中表达阳性率为90%(18/20),癌旁组织hMLH1表达明显高于癌组织有10例。其中有淋巴结转移的癌组织中hMLH1的表达缺失率为40%(4/10),无淋巴结转移的为50%(5/10);肿瘤浸润至外膜层的表达缺失率为40%(4/10)。结论 hMLH1基因在转录水平有异常表达,提示其在维吾尔族食管癌的发生、发展过程中起一定的作用。
Objective To investigate the expression of hMLH1 gene in esophageal carcinoma of Uighur in Xinjiang and its role in the development and progression of esophageal carcinoma. Methods RT-PCR was used to detect the expression of hMLH1 gene in 20 cases of esophageal cancer tissues and corresponding paracancerous tissues, and its significance was analyzed by clinicopathological data. Results The positive expression rate of hMLH1 was 55% (11/20) in esophageal cancer tissues, 90% (18/20) in paracancerous tissues and hMLH1 in paracancerous tissues was significantly higher than that in 10 cases. Among them, the rate of hMLH1 expression loss was 40% (4/10) and no lymph node metastasis was 50% (5/10) in cancerous tissues with lymph node metastasis. The rate of expression of hMLH1 in tumor invasion to epicardium was 40% (4 / 10). Conclusion The hMLH1 gene is abnormally expressed at the transcriptional level, suggesting that it plays a role in the occurrence and development of Uighur esophageal cancer.
