老年性疾呆病人基底前脑和纹状体内乙酰胆碱阳性神经元减少,活性 降低.纹状体内的胆碱能神经元和老年性遗忘有关,注射拟胆碱能药物能改善学习记忆功能.纹状体边缘区是我们在纹状体内发现的一个新区,含有大量的胆碱能纤维及神经元.边缘区的神经元发出纤维投射到与学习记忆功能有密切关系的Meynert基底核,并与其胆碱能阳性神经无形成突触.迷宫实验证明损伤两侧纹状体边缘区后动物的学习记忆能力显著下降.为进一步研究纹状体边缘区的神经元是否接受脑内其它部位传来的胆碱能投射,有无相应的胆碱能受体,以阐明纹状体边缘区学习记忆功能的机理,以及纹状体边缘区在脑的学习记忆神经网络中的地位.本研究用地高辛精标记的nAChR cDNA探针进行原位杂交,观察nAChR mRNA在纹状体内的分布情况.结果在纹状体内观察到大量的nAChR mRNA表达.nAChR mRNA阳性反应产物呈深蓝色或紫蓝色颗粒状,分布在细胞体及树突基部的细胞膜上,纹状体尾壳核中阳性细胞较多,为中等大的多极形细胞.苍白球内只有少量大的阳性多极形细胞.边缘区内则可见较多的nAChR mRNA阳性细胞,为中等大的梭形细胞,呈密集的带状分布.本结果提示纹状体边缘区内有较多的nAChR mRNA表达,接受来自脑内其它部位胆碱能纤维的传人
