目的:采用超高效液相色谱法建立降脂抗氧化合剂中葛根素和2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量测定方法。方法:色谱柱:ACQUITY UPLC BEH C18(100 mm×2.1 mm,1.7μm);流动相:乙腈与0.1%甲酸溶液,梯度洗脱;检测波长:305 nm;流速:0.4 mL/min;柱温:30℃;进样量:2μL。结果:葛根素的线性范围为0.02~1.0 mg/mL,r1=0.9998,平均回收率为98.58%,RSD为1.8%;2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的线性范围为0.005~0.2 mg/mL,r2=0.9990,平均回收率为99.62%,RSD为1.2%。结论:该方法简便、快速、准确,可用于降脂抗氧化合剂的质量控制。
OBJECTIVE: To establish a method for the determination of puerarin and 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside in lipid-lowering anti-oxidative agents by ultra performance liquid chromatography. Method: Column: ACQUITY UPLC BEH C18 (100 mm × 2.1 mm, 1.7 μm); mobile phase: gradient elution with acetonitrile and 0.1% formic acid; detection wavelength: 30 ℃; injection volume: 2μL. Results: The linear range of puerarin was 0.02-1.0 mg / mL, r1 = 0.9998, the average recovery was 98.58% and the RSD was 1.8%. The concentration of 2,3,5,4’-tetrahydroxystilbene-2-O- The linear range of β-D-glucoside was 0.005-0.2 mg / mL, r2 = 0.9990, the average recovery was 99.62% and the RSD was 1.2%. Conclusion: The method is simple, rapid and accurate and can be used for the quality control of lipid-lowering antioxidants.
