目的研究燕麦β-葡聚糖对高脂血症大鼠肝损伤的保护作用。方法健康成年SD大鼠70只,雌雄各半,随机分成正常对照组(n=10)和模型组(n=60)。模型组喂高脂饲料,3w后通过对实验大鼠进行血脂检测、B超检查和随机挑选性解剖,确定模型组的肝损伤。挑选50只肝损伤大鼠,按血脂随机分为5组(n=10):燕麦β-葡聚糖低、中、高剂量组[133、266和533 mg/(kg bw.d),灌胃],高脂对照组(灌胃等量蒸馏水),降脂对照组[壳寡糖200 mg/(kg.d),灌胃]。正常对照组喂普通饲料、蒸馏水4 w,其它各组继续喂高脂饲料4 w。分别于大鼠灌胃给药前及给药4 w眼眶采血,测定血清ALT、AST和MDA;末次采血后取大鼠肝脏标本并检测其肝脏MDA指标。结果与高脂对照组相比,燕麦β-葡聚糖高剂量组的血清ALT、AST明显降低,血清及肝脏MDA水平下降最明显,中剂量组和壳寡糖组MDA下降水平次之;低剂量组下降无统计学意义。三个剂量组肝细胞变性、坏死明显轻于对照组。结论燕麦β-葡聚糖对大鼠高脂血症所造成的肝损伤有保护作用。
Objective To study the protective effect of oat β-glucan on liver injury in hyperlipidemic rats. Methods Seventy healthy adult SD rats were randomly divided into normal control group (n = 10) and model group (n = 60). The model group was fed with high-fat diet. After 3 weeks, the rats in the model group were subjected to the blood lipids test, B-ultrasound examination and randomly selected anatomy to determine the liver injury in the model group. Fifty rats with liver injury were selected and randomly divided into five groups (n = 10): lipoprotein β-glucan low, medium and high dose groups [133,266 and 533 mg / (kg bw.d) Stomach], high-fat control group (intragastric administration of distilled water equivalent), lipid-lowering control group [oligochitosan 200 mg / (kg.d), intragastric administration]. Normal control group fed normal feed, distilled water 4 w, the other groups continue to feed high fat diet 4 w. Serum ALT, AST and MDA were measured before and 4 weeks after oral administration in rats, respectively. The liver samples were taken from the rats after the last blood collection and the liver MDA contents were measured. Results Compared with the high-fat control group, the levels of serum ALT and AST were significantly decreased and the levels of MDA in serum and liver were significantly decreased in the oat β-glucan high-dose group, followed by MDA in the medium-dose and oligochitosan groups There was no statistical significance in the dose group. Three doses of liver degeneration, necrosis was significantly lighter than the control group. Conclusion oat β-glucan has a protective effect on liver injury induced by hyperlipidemia in rats.
