目的 本研究旨在比较靶向bcl- 2基因的反义mRNA和dsRNA对SP2 O细胞生长的抑制作用。方法 采用MTT法,免疫荧光和免疫细胞化学技术,克隆形成试验及分光光度计测定细胞总蛋白含量等方法,分析比较了经体外转录获得的bcl- 2反义mRNA利LdsRNA抑制SP2 O细胞生长、bcl- 2特异性蛋白和总蛋白表达及其克隆原性的作用。结果 检测结果证实bcl- 2反义mRNA和dsRNA对上述指标均有抑制作用,两者比较dsRNA的抑制作用更强,持续时间也较长,作为对照的bcl- 2正义RNA对上述指标无显著增强作用。提示LdsRNA对bcl- 2高表达的肿瘤细胞有更强的抑制和杀伤作用,因此有可能用于这些肿瘤的临床治疗。
Purpose This study aimed to compare the inhibitory effects of antisense mRNA and dsRNA targeting the bcl-2 gene on SP2 O cell growth. Methods MTT assay, immunofluorescence and immunocytochemistry, clonogenic assay and spectrophotometer determination of total cellular protein content and other methods were analyzed by in vitro transcription of bcl-2 antisense mRNA and LdsRNA inhibit SP2 O cell growth, The role of bcl-2-specific protein and total protein expression and their clonogenicity. Results The results showed that both bcl-2 antisense mRNA and dsRNA had inhibitory effect on the above indexes, both of them had stronger inhibitory effect and longer duration compared with dsRNA, and the bcl-2 sense RNA as a control had no significant enhancement on the above indexes effect. Tip LdsRNA on bcl-2 high expression of tumor cells have a stronger inhibitory and killing effect, it is possible for the clinical treatment of these tumors.
