目的研究线粒体ATP敏感性钾通道开放剂二氮嗪对异丙肾上腺素诱导的大鼠心肌细胞凋亡相关基因表达的影响。方法采用完全随机化设计,将32只雄性SD大鼠随随机分为4组:A.正常对照组;B.异丙肾上腺素组;C.二氮嗪组;D.格列本脲组。采用半定量RT-PCR法检测各组大鼠心肌Bcl-2、Caspase-3相对表达量;采用TUNEL法检测各组大鼠心肌凋亡,计算凋亡指数(apopticindex,AI)。结果各组大鼠心肌Bcl-2、Caspase-3相对表达量之间存在差异(P<0.01)。C组Bcl-2、Caspase-3相对表达量与A,B,D组均有差异(P<0.05);B组、D组与A组之间有差异(P<0.05);B组、D组之间无明显差异(P=0.134,0.297)。A组大鼠心肌TUNEL染色未见阳性细胞;B、B、D各组AI上存在差异(P=0.001);C组和B、D组织间存在差异(P<0.01);B组、D组之间无明显差异(P=0.156)。结论二氮嗪可以减少异丙肾上腺素诱导的大鼠心肌细胞凋亡;减少凋亡的原因可能与其能上调Bcl-2的表达,下调Caspase-3表达有关。
Objective To investigate the effects of mitochondrial ATP-sensitive potassium channel opener diazoxide on isoproterenol-induced apoptosis-related gene expression in rat cardiomyocytes. Methods A total of 32 male Sprague-Dawley rats were randomly divided into 4 groups according to a completely randomized design: A. normal control group; B. isoproterenol group; C. diazoxide group; D. glyburide group. The relative expression of Bcl-2 and Caspase-3 in each group was detected by semi-quantitative RT-PCR. Apoptosis was detected by TUNEL method and the apoptic index (AI) was calculated. Results The relative expressions of Bcl-2 and Caspase-3 in myocardium of rats in each group were different (P <0.01). The relative expression of Bcl-2 and Caspase-3 in group C were significantly different from those in groups A, B and D (P <0.05); there was a significant difference between group B and group D (P <0.05) There was no significant difference between groups (P = 0.134, 0.277). There were no positive cells in TUNEL staining in group A, there were differences in AI in groups B, B and D (P = 0.001), there was a significant difference between groups C and B and D (P <0.01) No significant difference (P = 0.156). Conclusions Diazoxide can reduce isoproterenol-induced apoptosis in rat cardiomyocytes. It may be related to its role in up-regulating the expression of Bcl-2 and down-regulating the expression of Caspase-3.
