目的观察β-catenin短发夹RNA干扰(shRNA)重组慢病毒阻断Wnt/β-catenin信号通路及其对染尘小鼠肺泡灌洗液中细胞因子表达的影响。方法将小鼠分为生理盐水组、silica+Lv-shβ-catenin组、silica模型组(1mg Si O2/小鼠)和silica+Lv-shβ-catenin-NC组。染尘后7,28,56天处死动物,收集肺泡灌洗液。分别采用Western Blot和ELISA方法测定小鼠肺组织中β-catenin蛋白表达和肺泡灌洗液中白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达情况。结果染尘后各时间点,silica+Lv-shβ-catenin组小鼠肺组织中β-catenin蛋白表达均低于silica组和silica+Lv-shβ-catenin-NC组;且silica+Lv-shβ-catenin组小鼠肺泡灌洗液中IL-1β、IL-6和TNF-α表达均显著低于silica组和silica+Lv-shβ-catenin-NC组,且差异有统计学意义(P<0.05)。结论通过干扰染尘小鼠肺组织中β-catenin的表达能够有效抑制其肺泡灌洗液中IL-1β、IL-6和TNF-α的表达。
Objective To observe the effect of short interfering RNA (shRNA) recombinant lentivirus of β-catenin on the Wnt / β-catenin signaling pathway and its effect on the expression of cytokines in the alveolar lavage fluid of mice exposed to β-catenin. Methods The mice were divided into normal saline group, silica + Lv-shβ-catenin group, silica model group (1 mg Si O2 / mouse) and silica + Lv-shβ-catenin-NC group. The animals were killed 7, 28, 56 days after the dust was collected and the alveolar lavage fluid was collected. The expression of β-catenin and IL-1β, IL-6 and TNF-α in bronchoalveolar lavage fluid were detected by Western Blot and ELISA respectively. α) expression. Results The expression of β-catenin in the lung tissue of silica + Lv-shβ-catenin group was lower than that in silica group and silica + Lv-shβ-catenin-NC group at each time point after exposure to dust. The expression of IL-1β, IL-6 and TNF-α in bronchoalveolar lavage fluid of catenin group was significantly lower than that of silica group and silica + Lv-shβ-catenin-NC group (P <0.05) . Conclusion The expression of β-catenin in lung tissue of mice with dust-induced lung injury can effectively inhibit the expression of IL-1β, IL-6 and TNF-α in BALF.
