以伴刀豆凝集素A刺激小鼠脾细胞的条件培养基作为巨核细胞集落刺激因子(CSF-MK)的来源,研究小鼠骨髓巨核祖细胞在体外培养中的增殖和分化。骨髓细胞经9d培养结果表明,巨核细胞的数量在培养第3d开始增高,5~7d增殖达到高峰,之后逐渐下降;在培养的同一时间内,还可观察到分化程度不同的巨核细胞,提示巨核祖细胞是具有年龄结构的细胞群及CFU-Meg细胞池具有不均一性的特征。
To investigate the proliferation and differentiation of mouse bone marrow megakaryocyte progenitor cells in vitro, the conditioned medium of spleen cells stimulated by Concanavalin A was used as the source of megakaryocyte colony-stimulating factor (CSF-MK). The results of 9d culture showed that the number of megakaryocytes began to increase from the 3rd day of culture to the 5th day, reached the peak at 5 ~ 7d, and then gradually decreased. At the same time, megakaryocytes with different degrees of differentiation were also observed, Progenitor cells are characterized by heterogeneity of age-structured cell populations and CFU-Meg cell pools.
