目的探讨小剂量地塞米松对急性呼吸窘迫综合征(ARDS)大鼠的保护作用及对糖皮质激素受体表达的影响。方法油酸复制ALI/ARDS大鼠模型。随机将18只SD大鼠分为三组:对照组(A1)、ARDS组(A2)、地塞米松组(A3),每组6只。测各组大鼠动脉氧分压,肺组织病理切片,ELISA法测各组大鼠血液中TNF-α,免疫组化法测肺组织NF-ΚBp65,荧光定量RCR测各组糖皮质激素受体(GR)表达量。结果各组之间病理改变有明显差异,以下各指标:氧分压,TNF-α,肺组织NF-κBp65含量,糖皮质激素受体(GR)表达量,各组之间差异均有统计学意义(P<0.05)。结论小剂量地塞米松对ARDS大鼠肺组织有保护作用,其作用机制可能与对糖皮质激素受体起到正向调节作用有关。
Objective To investigate the protective effect of low dose dexamethasone on acute respiratory distress syndrome (ARDS) rats and its effect on glucocorticoid receptor expression. Methods Oleic acid replication in ALI / ARDS rat model. Eighteen SD rats were randomly divided into three groups: control group (A1), ARDS group (A2), dexamethasone group (A3), 6 rats in each group. The arterial oxygen pressure and the histopathological sections of the lungs of rats in each group were measured. The levels of TNF-α in the blood of the rats in each group were measured by ELISA. The expressions of NF-κBp65 in the lung tissue were detected by the immunohistochemical method. The expressions of glucocorticoid receptor (GR) expression level. Results There were significant differences in the pathological changes between the groups. The following indexes were observed: oxygen partial pressure, TNF-α, NF-κBp65 content and glucocorticoid receptor (GR) expression in each group Significance (P <0.05). Conclusion Low-dose dexamethasone has a protective effect on the lung tissue of ARDS rats, and its mechanism may be related to the positive regulation of glucocorticoid receptor.
