目的观察不同浓度肿瘤坏死因子相关凋亡诱导配体(TRAIL)对宫颈癌HeLa细胞的凋亡作用。方法2006年于重庆医科大学,采用不同浓度的TRAIL作用于HeLa细胞,采用MTT(噻唑蓝)法检测细胞存活分数;AO/EB(丫碇橙/嗅乙啶)双重荧光染色观察凋亡细胞结构;流式细胞仪检测细胞周期及细胞凋亡率。结果TRAIL作用于HeLa细胞24h后,TRAIL对细胞生长抑制率及细胞凋亡率具有量效关系;AO/EB双重荧光染色可见到早期、晚期凋亡细胞;流式细胞仪检测有亚二倍峰出现;细胞周期发生改变。结论TRAIL对HeLa细胞生长有抑制作用,并可诱导其凋亡。
Objective To observe the apoptosis effect of different concentration of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) on cervical cancer HeLa cells. Methods In Chongqing Medical University in 2006, different concentrations of TRAIL were applied to HeLa cells. MTT (thiazolyl blue) method was used to detect cell viability. AO / EB staining was used to observe the apoptotic cell structure Flow cytometry was used to detect cell cycle and apoptosis rate. Results After treated with TRAIL for 24 h, TRAIL had dose-response relationship with cell growth inhibition rate and apoptosis rate. Apoptotic cells in early and late stage were observed by AO / EB double staining. Flow cytometry showed sub-double peak Appear; the cell cycle changes. Conclusion TRAIL can inhibit HeLa cell growth and induce apoptosis.
