甲基莲心碱(Nef)给药后5min使豚鼠离体工作心脏的左室压(LVP)下降17.9%(P<0.01),左室压瞬时变化速率(±dp/dt_(max))分别下降22.6%和11.7%(P均<0.05),左室舒末压(LVEDP)上升48.6%(P<0.01),主动脉流量(ABF)减少48.4%(P<0.01),冠脉流量(CBF)增加10.7%(P<0.05),心率(HR)变化不明显,Nef0.1~10.0μmol/L剂量依赖性地降低LVP、±dP/dt_(max)、ABF,而CBF在较低剂量范围内是增加的。Nef对离体豚鼠工作心脏各参数的影响类似维拉帕米。结果提示,Nef对心脏收缩性能呈剂量依赖性抑制。减少ABF,而在一定剂量范围内可增加冠脉流量。
