目的:观察银杏叶提取物(GbE)对肝纤维大鼠肝组织环氧合酶-2(COX-2)、Ⅰ型胶原表达的影响,探讨其抗肝纤维化的作用机制。方法:采用40%的CCl4皮下注射6周建立大鼠肝纤维化模型,以200、100、50mg.kg-1.d-1的GbE干预,原位杂交检测肝组织COX-2mRNA表达,免疫组化检测肝组织Ⅰ型胶原、COX-2蛋白表达。结果:模型组大鼠肝组织Ⅰ型胶原蛋白、COX-2mRNA和蛋白表达均较正常组明显增强,应用GbE干预后,大鼠Ⅰ型蛋白、COX-2mRNA和蛋白表达均明显减少。结论:GbE抗肝纤维化作用的机制之一可能与其下调肝组织中COX-2的表达,减少胶原的产生,改善肝脏微循环有关。
Objective: To observe the effect of Ginkgo biloba extract (GbE) on the expression of cyclooxygenase-2 (COX-2) and collagen Ⅰ in hepatic tissues of rats with hepatic fibrosis and to explore its mechanism of action against hepatic fibrosis. Methods: The model of hepatic fibrosis was established by subcutaneous injection of 40% CCl4 for 6 weeks. The expression of COX-2 mRNA in liver tissue was detected by GbE intervention at 200, 100 and 50 mg.kg-1.d-1, To detect the expression of type Ⅰ collagen and COX-2 protein in liver tissue. Results: The expressions of type Ⅰ collagen and COX-2 mRNA and protein in the model group were significantly increased compared with those in the normal group. The expressions of type Ⅰ protein and COX-2 mRNA and protein in the model group were significantly decreased after GbE intervention. CONCLUSION: One of the mechanisms of anti-hepatic fibrosis in GbE may be related to down-regulation of COX-2 expression in liver tissue, reduction of collagen production and improvement of hepatic microcirculation.
