目的:研究K562/A02细胞株NF-кB活性与P-gp、Bcl-2表达的关系。方法:在K562/A02白血病耐药细胞株培养中加入NF-кB抑制剂PDTC,观察在不同浓度、不同时间下,细胞株NF-кB活性的变化与P-gp、Bcl-2表达的关系。结果:在不同浓度、不同时间的PDTC下,K562/A02细胞株NF-кB活性与P-gp、Bcl-2表达呈正相关的关系,随干预浓度、时间的不断变化,三者均逐渐降低。结论:NF-кB调控的白血病细胞抗凋亡机制中有MDR1、Bcl-2的参与。
Objective: To study the relationship between the activity of NF-κB and the expressions of P-gp and Bcl-2 in K562 / A02 cell line. Methods: NF-кB inhibitor PDTC was added into K562 / A02 leukemia cell line to observe the relationship between the changes of NF-кB activity and the expressions of P-gp and Bcl-2 under different concentrations and different time. RESULTS: The NF-кB activity of K562 / A02 cell line was positively correlated with the expression of P-gp and Bcl-2 under different concentrations of PDTC. With the concentration and time changing constantly, all three showed a decreasing trend. CONCLUSION: The anti-apoptotic mechanisms of NF-кB in leukemia cells involve MDR1 and Bcl-2.
