目的观察癫痫发作后大鼠脑组织中 Fas的表达及细胞凋亡的情况以及神经生长因子 (NGF)的干预性作用。方法采用 LiCL- Pilocarpin致痫的 Wister大鼠模型,通过 TUNEL和免疫组化方法对各组 Fas的表达、细胞凋亡情况进行观察。结果癫痫后 1hFas表达开始增加, 12 h达高峰,持续 1周。 Fas的高表达与细胞凋亡的分布一致。 NGF组 Fas表达和细胞凋亡情况均较致痫组有显著的减少 (P< 0.01)。结论癫痫发作可引起 Fas表达和细胞凋亡增加。 NGF有抑制 Fas表达和细胞凋亡增加的作用。
Objective To observe the expression of Fas and apoptosis in rat brain after epileptic seizure and the intervention of nerve growth factor (NGF). Methods Wister rat models of epilepsy induced by LiCL-Pilocarpin were used. TUNEL and immunohistochemical methods were used to observe the expression of Fas and apoptosis in each group. Results The expression of Fas began to increase 1 h after epilepsy and peaked at 12 h for 1 week. The high expression of Fas is consistent with the distribution of apoptosis. The expression of Fas in the NGF group was significantly lower than that in the epileptogenic group (P <0.01). Conclusion Epileptic seizures can induce Fas expression and increase apoptosis. NGF has the effect of inhibiting Fas expression and increasing apoptosis.
