应用甘油低渗两相分配法分离了SRSV/3T3细胞质膜(SM)和NIH/3T3细胞质膜(NM)。经Na~+、K~+-ATP酶和葡萄糖-6-磷酸酶等标志酶活性鉴定,表明SM和NM纯度较高。经SDS-PAGE分析比较,发现SM和NM的蛋白质组分有较明显差异。SM有7种独特的组分,分子量分别为2.29×10~(-22)、2.18×10~(-22)、9.79×10~(-23)、9.60×10~(-23)、8.63×10~(-23)、5.49×10~(-23)和4.78×10~(-23)kg。NM有两种特有的组分(1.15×10~(-22)和4.17×10~(-23)kg)。在彼此共有的蛋白质组分中,也有许多组分在含量和电泳迁移速度上存在着程度不同的区别。
SRSV / 3T3 plasma membrane (SM) and NIH / 3T3 plasma membrane (NM) were isolated by glycerol hypotonic two-phase partitioning. The enzyme activity of Na ~ +, K ~ + -ATPase and glucose-6-phosphatase and other markers identified, indicating that SM and NM purity higher. By SDS-PAGE analysis and comparison, found that SM and NM protein components have more significant differences. SM has seven unique components with molecular weights of 2.29 × 10 ~ (-22), 2.18 × 10 ~ (-22), 9.79 × 10 ~ (-23), 9.60 × 10 ~ (-23), 8.63 × 10 ~ (-23), 5.49 × 10 ~ (-23) and 4.78 × 10 ~ (-23) kg, respectively. NM has two peculiar components (1.15 × 10 ~ (-22) and 4.17 × 10 ~ (-23) kg). Among the protein components shared with each other, there are also many components that differ in their contents and the electrophoretic mobility to some extent.
