目的:研究吴茱萸碱和吴茱萸次碱微透析探针体外回收率及传递率与流速、药物浓度之间的关系。方法:浓差法(正、反向透析法)考察流速及药物浓度对微透析探针的体外回收率和传递率的影响,以HPLC法测定微透析样品中EVO、RUT浓度。结果:探针体外回收率与传递率基本一致,均与灌流速度成反比,药物浓度对其影响不大。结论:在吴茱萸碱和吴茱萸次碱的药动学研究中,可采用反透析法作为探针回收率的测定方法。
OBJECTIVE: To study the relationship between evodiamine and rutaecarpine microdialysis probe in vitro recovery, transudation rate, flow rate and drug concentration. Methods: Concentration difference method (forward and reverse dialysis) was used to investigate the effect of flow rate and drug concentration on microdialysis probe recovery rate and transmissibility in vitro. The concentrations of EVO and RUT in microdialysis samples were determined by HPLC. Results: The recovery rate and transmissibility of probe in vitro were basically the same, all of them were inversely proportional to the perfusion rate, and the effect of drug concentration was insignificant. CONCLUSION: In the pharmacokinetic studies of evodiamine and rutaecarpine, reverse dialysis may be used as a method for the determination of probe recovery.
