目的:观察雷公藤内酯醇是否能够影响淋巴细胞内环磷酸腺苷(cAMP)水平及依赖cAMP蛋白激酶(PKA),探讨雷公藤内酯醇影响T细胞功能的作用机制。方法:以人外周血T细胞为研究对象,分别采用放射免疫法和酶底物显色法测定细胞内cAMP水平及PKA酶活性。结果:(1)雷公藤内酯醇能够明显增加T细胞内cAMP水平,同时细胞内PKA酶活性也明显增强,并且与雷公藤内酯醇的浓度密切相关;(2)应用腺苷酸环化酶抑制剂SQ22536,能够抑制雷公藤内酯醇所致的细胞内cAMP水平升高和PKA活性增强;(3)cAMP和PKA拮抗剂能够拮抗雷公藤内酯醇升高细胞cAMP水平及增强PKA活性的作用。结论:雷公藤内酯醇能够通过影响腺苷酸环化酶活性,在增加T细胞内cAMP水平的同时增强细胞内PKA活性
Objective: To observe whether triptolide can affect the level of cAMP in lymphocytes and the cAMP-dependent protein kinase (PKA), and to explore the mechanism of triptolide affecting T cell function. Methods: Human peripheral blood T cells were used as the research object. The intracellular cAMP level and PKA enzyme activity were determined by radioimmunoassay and enzyme substrate colorimetry respectively. Results: (1) Triptolide could significantly increase the level of cAMP in T cells, and the activity of PKA in cells was also significantly increased, and it was closely related to the concentration of triptolide; (2) Application of adenylate cyclase inhibition SQ22536 can inhibit the increase of intracellular cAMP level and PKA activity induced by triptolide. (3) cAMP and PKA antagonists can antagonize the effects of triptolide on elevating cellular cAMP levels and enhancing PKA activity. Conclusion: Triptolide can affect the activity of adenylyl cyclase and increase the intracellular cAMP level in T cells while increasing the intracellular PKA activity.
