目的:探讨B7同源体4(B7 homolog 4,B7-H4)在人乳腺癌组织的表达及其与肿瘤内CD4~+T、CD8~+T细胞的数量和IFN-γ细胞因子分泌的关系。方法:应用S-P免疫纽织化学染色检测B7-H4在乳腺癌、癌旁组织和乳腺良性病变组织中的表达;间接免疫荧光双标染色分析乳腺癌组织内浸润的CD4~+T、CD8~+T细胞数量及TGF-β1和IFN-γ分泌情况。结果:B7-H4在乳腺癌、乳腺良性病变和癌旁组织的阳性表达率分别为75.0%、37.5%和25.0%,三组间存在显著性差异(均P<0.01);B7-H4阳性乳腺癌组织内浸润的CD8~+T细胞及IFN-γ分泌细胞数量明显低于B7-H4阴性的乳腺癌组织(P<0.05),但B7-H4的表达与乳腺癌组织内浸润的CD4~+T细胞及TGF-β1分泌细胞数量无关。结论:B7-H4在乳腺癌中表达明显上调,并与抑制CD8~+T细胞浸润和分泌IFN-γ有关。B7-H4在下调乳腺癌抗肿瘤细胞免疫中具有一定作用。
Objective: To investigate the expression of B7 homolog 4 (B7-H4) in human breast cancer and its relationship with the number of CD4 ~ + T, CD8 ~ + T cells and the secretion of IFN-γ in tumor tissue . Methods: Immunohistochemical staining was used to detect the expression of B7-H4 in breast cancer tissues, paracancerous tissues and benign breast lesions. Indirect immunofluorescence double staining was used to detect the infiltration of CD4 ~ + T and CD8 ~ + in breast cancer tissues. T cells and TGF-β1 and IFN-γ secretion. Results: The positive rates of B7-H4 in breast cancer, benign breast lesions and adjacent non-cancerous tissues were 75.0%, 37.5% and 25.0%, respectively (all P <0.01) The number of infiltrating CD8 ~ + T cells and IFN-γ secreting cells in cancerous tissues was significantly lower than that in B7-H4-negative breast cancers (P <0.05), but the expression of B7-H4 was significantly correlated with infiltrating CD4 ~ T cells and TGF-β1 secretion of cells has nothing to do. Conclusion: The expression of B7-H4 in breast cancer is significantly up-regulated, and it is related to the inhibition of CD8 + T cell infiltration and secretion of IFN-γ. B7-H4 in breast cancer down-regulation of anti-tumor cell immunity has a role.
