目的研究太行菊Opisthopappus taihangensis和野菊花Dendranthema indicum不同部位、不同浓度水提取物对乙型肝炎病毒(HBV)的抑制作用。方法提取并配置不同浓度的太行菊和野菊花的花和茎部位的水提取液,分别以拉米夫定(3TC)和绿茶提取物(GTE)作对照,采用酶联免疫反应检测太行菊花和茎提取物对HBs Ag、HBe Ag的抑制作用,采用实时定量PCR检测不同浓度提取物对HBV复制过程中HBV RNA的影响,荧光素酶报告基因检测不同提取物对HBV转录的影响。结果酶联免疫反应结果表明,1μg/m L太行菊茎提取物能够在作用48、96 h时抑制HBs Ag和HBe Ag,而1μg/m L野菊花茎提取物只能在作用48 h时抑制HBs Ag和HBe Ag;0.1μg/m L太行菊花提取物与1μg/m L野菊花花提取物对HBs Ag和HBe Ag的抑制作用相当。荧光素酶报告基因检测结果说明,太行菊花和茎提取物相对野菊花和茎提取物对HBV启动子质粒CP有抑制作用。PCR结果显示,太行菊花和茎提取物在低浓度时对HBV RNA合成具有一定的抑制作用。结论太行菊水提取物和野菊花提取物都能够不同程度抗HBV,但太行菊效果优于野菊花。
Objective To study the inhibitory effect of water extract of different concentrations on the Hepatitis B virus (HBV) in different parts of Opisthopappus taihangensis and Dendranthema indicum. Methods The aqueous extracts of flowers and stems from different concentrations of Taihang chrysanthemum and Chrysanthemum morifolium were extracted and equipped with 3TC and GTE as control respectively. Enzyme-linked immunosorbent assay Stem extract on HBsAg, HBeAg, using real-time quantitative PCR detection of different concentrations of extracts on HBV RNA replication during HBV RNAi, luciferase reporter gene detection of different extracts on HBV transcription. Results The results of enzyme-linked immunosorbent assay showed that 1μg / m L Taihanguang stem extract could inhibit HBsAg and HBeAg at 48 and 96 hours, while 1μg / m L wild chrysanthemum stem extract could only inhibit HBs Ag and HBe Ag; 0.1μg / m L Taihang Chrysanthemum extract and 1μg / m L Chrysanthemum flower extract on HBsAg and HBeAg inhibition. Luciferase reporter gene test results show that the Taihang chrysanthemum and stem extract relative wild chrysanthemum and stem extract on the HBV promoter plasmid CP inhibition. PCR results showed that the extract of Taihang chrysanthemum and stem had a certain inhibitory effect on HBV RNA synthesis at low concentration. Conclusion Taihang Jushui extract and wild chrysanthemum extract all have anti-HBV to varying degrees, but Taihang Ju better than wild chrysanthemum.
