目的探讨钙蛋白酶(calpain 1)在卡那霉素(kanamyc in,KM)致毒豚鼠耳蜗中的定位与表达。方法将40只豚鼠随机均分成4组:对照组、KM 3 d组、KM 7 d组和KM 14 d组,KM各组每天肌肉注射硫酸卡那霉素注射液200 mg·kg-1。应用电生理指标听性脑干反应(aud itory brainstem response,ABR)观察用药前后豚鼠听力的变化;采用免疫组织化学SABC法结合显微图像分析技术,观察豚鼠耳蜗组织中calpain 1的表达。结果 calpain 1阳性免疫反应主要见于耳蜗毛细胞、螺旋神经节、螺旋韧带和血管纹,并随给药天数的增加,calpain 1在耳蜗上述部位的表达逐渐减弱。结论 calpain 1在正常豚鼠和卡那霉素致毒豚鼠耳蜗中都有表达,且随卡那霉素给药天数的增加表达逐渐减弱。
Objective To investigate the localization and expression of calpain 1 in the guinea pig cochlea induced by kanamyc in (KM). Methods Forty guinea pigs were randomly divided into four groups: control group, KM 3 d group, KM 7 d group and KM 14 d group. KM groups were intramuscularly injected with kanamycin sulfate 200 mg · kg -1 every day. The auditory brainstem response (ABR) was used to observe the changes of auditory brainstem response in guinea pigs. The expression of calpain 1 in guinea pig cochleas was observed by immunohistochemical SABC combined with microscopic image analysis. Results The positive immunoreactivity of calpain 1 was mainly found in cochlear hair cells, spiral ganglion, spiral ligament and vascular pattern. The expression of calpain 1 in the above cochlea gradually decreased with the increase of dosing days. Conclusion calpain 1 is expressed in the cochlea of normal guinea pigs and kanamycin-induced guinea pigs, and gradually decreases with the increase of kanamycin-administered days.
