目的研究脑缺血再灌注损伤大鼠脑组织中Livin和Survivin的表达及氟伐他汀对其表达的影响。方法用线栓法制作大鼠脑缺血再灌注模型。大鼠随机分为假手术组、模型组和实验组,每组8只。实验组在模型制作前予以10mg·kg~(-1)·d~(-1)氟伐他汀,连续灌胃14 d;假手术组和模型组正常喂养。于缺血2 h再灌注24 h后,用免疫组织化学法、半定量聚合酶链式反应(PCR)法和免疫印迹法检测脑组织中Livin和Survivin的表达情况。结果与假手术组相比,模型组Livin和Survivin表达均显著增高,差异均有统计学意义(P<0.05)。与模型组相比,实验组Livin和Survivin表达均进一步显著增高,差异均有统计学意义(P<0.05)。结论氟伐他汀可通过增强Livin和Survivin表达抵制细胞凋亡,从而发挥脑保护作用。
Objective To investigate the expression of Livin and Survivin in rat brain with cerebral ischemia-reperfusion injury and the effect of fluvastatin on it. Methods The rat model of cerebral ischemia-reperfusion was established by thread occlusion. Rats were randomly divided into sham operation group, model group and experimental group, with 8 rats in each group. The experimental group was treated with fluvastatin 10 mg · kg -1 d -1 for 14 days before the model was established. The sham operation group and the model group were fed with normal saline. After reperfusion for 2 h at 2 h of ischemia, the expression of Livin and Survivin in brain tissue were detected by immunohistochemistry, semi-quantitative polymerase chain reaction (PCR) and Western blotting. Results Compared with the sham operation group, the expression of Livin and Survivin in the model group were significantly increased, the differences were statistically significant (P <0.05). Compared with the model group, the expression of Livin and Survivin in the experimental group were further significantly increased, the differences were statistically significant (P <0.05). Conclusions Fluvastatin can protect brain cells by increasing the expression of Livin and Survivin.
