鉴于扁平苔癣损害中的炎症浸润在发病原因上的重要意义,作者用体外自动射线显影和电子显微镜研究口腔粘膜和皮肤扁平苔癣的炎症浸润的性质,其研究步骤为: (1)在治疗前切取14例颊粘膜损害的活检标本(年龄40—75岁,平均54岁),切取时间始终在下午3—5时以尽量减少标记活性在白昼的变异。每个标本分为两份,一半标本固定在10%福尔马林盐水中,供光学显微镜诊断之用,另一半切成立方形的小块,每边不能大于1毫米,供自动射线显影之用。这些小方块在含有10%胎牛血清和5微居里氚标记胸腺嘧啶的培养基内,在37℃下培育一小时,然后固定在10%福尔马林盐水中,按照Walker and Dolby(1974)氏法作自动射线显影。
