通过动态吸附和解吸附研究,从5种色谱填料中筛选出适合唾液酸乳糖与乳糖分离的色谱树脂,建立了唾液酸乳糖和乳糖的色谱分离技术,考察了径高比、洗脱速度、柱温、上样浓度等色谱条件对分离度和回收率的影响,确定了最佳色谱分离条件.结果表明,树脂QY-Z001可高效分离唾液酸乳糖和乳糖,最佳分离条件为径高比1:35、温度55℃、流速1.0 m L/min.、进样浓度100 g/L.该条件下唾液酸乳糖与乳糖分离度可达0.96,唾液酸乳糖含量由6.8%提高至84.6%,回收率为76.9%.
Chromatographic separation of sialyllactose and lactose was carried out by dynamic adsorption and desorption studies.
