目的:检测结肠直肠癌组织及相应癌旁组织中上皮细胞转化序列2癌基因(epithelial cell transforming sequence 2 oncogene,ECT2)的m RNA表达以及蛋白质表达,并探讨其与结肠直肠癌临床病理因素的相关性及临床意义。方法:采用实时定量聚合酶链反应法检测结肠直肠癌病人的癌组织和相应癌旁组织ECT2 m RNA的表达;免疫组织化学法检测150例癌组织ECT2蛋白表达。结果:结肠直肠癌组织ECT2 m RNA表达水平明显升高(P<0.001);而ECT2蛋白表达水平与肿瘤直径(P=0.005)、CEA(P=0.006)以及TNM分期(P=0.001)密切相关。Kaplan-Meier生存曲线和Log-Rank检验提示,ECT2表达水平越高,结肠直肠癌病人预后越差(P<0.001)。多因素COX回归分析表明,ECT2在结肠直肠癌病人中可作为独立的预后分子标志物。结论:ECT2 m RNA及蛋白质在结肠直肠癌组织中均呈高表达,且ECT2表达水平越高,病人预后越差。由此可见,ECT2在结肠直肠癌的发生、发展中起重要作用,可作为反映结肠直肠癌生物学行为的有效指标。
OBJECTIVE: To detect the expression of m RNA and protein in epithelial cell transforming sequence 2 oncogene (ECT2) in colorectal cancer tissues and corresponding paracancerous tissues, and to explore its correlation with clinical and pathological factors of colorectal cancer Sexual and clinical significance. Methods: The expression of ECT2 mRNA was detected by real-time quantitative polymerase chain reaction (RT-PCR) in 150 cases of colorectal cancer tissues and corresponding paracancerous tissues. Immunohistochemistry was used to detect the expression of ECT2 protein in 150 cases of cancer tissues. Results: The expression of ECT2 mRNA in colorectal cancer tissues was significantly increased (P <0.001), while the expression of ECT2 protein was closely related to tumor diameter (P = 0.005), CEA (P = 0.006) and TNM stage . Kaplan-Meier survival curves and Log-Rank test showed that the higher the level of ECT2 expression, the worse the prognosis of patients with colorectal cancer (P <0.001). Multivariate COX regression analysis showed that ECT2 could be used as an independent prognostic molecular marker in patients with colorectal cancer. Conclusion: ECT2 m RNA and protein are highly expressed in colorectal cancer tissues, and the higher the expression level of ECT2, the poorer the prognosis of patients. Thus, ECT2 plays an important role in the occurrence and development of colorectal cancer and can be used as an effective indicator of the biological behavior of colorectal cancer.
