目的:探讨骨髓单个核细胞(BMMNCs)移植治疗脑梗死大鼠的疗效及其脑内分化情况。方法:采用线栓法制备大脑中动脉梗死大鼠模型,筛选成功模型后随机分为溶剂组(vehicle组)和BMMNCs移植组(BMMNCs组),BMMNCs组于24h后经尾静脉注入1mLBrdu标记含1×107个BMMNCs细胞悬液,vehicle组注射同等剂量PBS溶液,采用神经功能损害严重程度评分(Nss)观察神经行为学功能,免疫荧光双标法示踪BMMNCs脑内分化情况。结果:与vehicle组相比,BMMNCs组Nss明显降低(P<0.05),BMMNCs组脑内梗死皮层区、海马齿状回检测到Brdu和Iba1双标阳性细胞。结论:BMMNCs移植在脑梗死大鼠脑内分化为小胶质样细胞,显著改善脑梗死大鼠神经行为学功能。
Objective: To investigate the therapeutic effect and the intra-brain differentiation of bone marrow mononuclear cells (BMMNCs) transplantation in rats with cerebral infarction. Methods: The rat model of middle cerebral artery occlusion (MCAO) was established by thread occlusion method. After the successful model was screened, the rats were randomly divided into solvent group (vehicle group) and BMMNCs transplantation group (BMMNCs group). BMMNCs group was injected with 1mL Brdu labeled 1 × 107 BMMNCs cell suspension. The vehicle group was injected with the same dose of PBS solution. Neurobehavioral function was observed by Nss. The differentiation of BMMNCs was observed by immunofluorescence double labeling. Results: Compared with vehicle group, Nss in BMMNCs group was significantly lower (P <0.05). Brdu and Iba1 double positive cells were detected in cerebral cortex and hippocampal dentate gyrus of BMMNCs group. Conclusion: BMMNCs transplanted into the brain of rats with cerebral infarction differentiated into microglia-like cells, which significantly improved the neurobehavioral function of rats with cerebral infarction.
