目的研究模拟高+G_x 环境中猴舌黏膜上皮热休克蛋白质70(HSP-70)的表达。方法以9只雄性猕猴为对象,随机分为4组,对照组+1 G_x/300 s;实验1、2、3组,分别为+15 G_x/200 s、+18 G_x/165 s、+21 G_x/140 s。采用免疫组织化学方法,研究模拟高+G_x 环境作用下猴舌黏膜上皮细胞组织病理学改变及 HSP-70基因的表达情况,探讨其表达水平的改变与高+G_x环境的关系。结果组织病理学观察:实验组和对照组猴舌黏膜上皮细胞无明显变化;免疫组织化学观察:对照组舌黏膜上皮细胞核 HSP-70呈阴性表达或弱阳性表达。实验组舌黏膜上皮细胞核HSP-70着色明显,呈强阳性表达,不同高+G_x 组之间无明显差别。结论本研究表明模拟高+G_x 环境可引起猴舌黏膜上皮细胞 HSP-70基因表达增强。
Objective To investigate the expression of heat shock protein 70 (HSP-70) in the mucosal epithelium of the tongue in simulated + G_x environment. Methods Nine male rhesus macaques were randomly divided into 4 groups: control group, 1 G_x / 300 s; experimental group 1, 2, and 3 were +15 G_x / 200 s, +18 G_x / 165 s, +21 G_x / 140 s. Immunohistochemistry was used to study the histopathological changes and HSP-70 gene expression in monkey tongue mucosa epithelial cells under simulated high + G_x conditions, and to explore the relationship between the changes of expression levels and the environment of high + G_x. Results Histopathological observation: There was no significant change in the goblet cells in the experimental group and the control group. Immunohistochemistry showed that the HSP47 expression in the control group was negative or weakly positive. In experimental group, HSP-70 staining of tongue mucosa epithelial cells was obvious and strongly positive. There was no significant difference between different high + G_x groups. Conclusion This study shows that simulated high + G_x environment can cause the expression of HSP-70 in monkey tongue epithelial cells.
