已发现一种新多醚抗生素Inostamycin可以逆转人癌KB-C4细胞的多重抗药性。它可以增强秋水仙碱对抗药细胞KB-C4的细胞毒性而不增强对原系细胞KB-3-1的作用。为研究多醚抗生素的作用机理,作者在本研究中观察了不同结构的多醚化合物对抗药KB-C4细胞的影响。将多重抗药性KB-C4细胞及其原系KB-3-1细胞分别与秋水仙碱和多醚抗生素共同培养,并用MTT检测法测定细胞生长情况,秋水仙碱对KB-C 4和KB-3-1的IC_(50)分别是5.9±1.9μg/ml、0.001 5±0.0012μg/ml,KB-C4对秋水仙碱的抗药
A new polyether antibiotic, Inostamycin, has been found to reverse the multi-drug resistance of human cancer KB-C4 cells. It enhances cytotoxicity of colchicine against drug-resistant cell KB-C4 without enhancing its effect on the original cell line KB-3-1. In order to study the mechanism of action of polyether antibiotics, we investigated the effect of polyether compounds of different structures on drug-resistant KB-C4 cells in this study. KB-C4 cells and their original KB-3-1 cells were co-cultured with colchicine and polyether antibiotics, and the cell growth was measured by MTT assay.
