以普通小麦细胞质雄性不育保持系 75 3369B为材料 ,分别用限制性内切酶EcoRⅠ和MboⅠ对 75 3369B的mtDNA进行完全酶切和部分酶切 ,构建了两个基因文库。其中用EcoRⅠ酶切得到的库 (以下简称ME库 )由 350个重组子构成 ;MboⅠ酶切所构建的库 (以下简称MM库 )由 4 50个重组子构成 ,MM库的库容量是 98 12 %。同时 ,讨论了两种建库方法的优缺点
In this study, two gene libraries were constructed by digesting and partially digesting the mtDNA of 753369B with restriction endonucleases EcoRⅠ and MboⅠ, respectively, using the maintainer line 75 3369B of common wheat. Among them, 350 recombinants were constructed by digestion with EcoRⅠ (hereinafter referred to as ME library); the MboⅠ digestion library (hereinafter referred to as MM library) consisted of 4 50 recombinants, and the MM library had a capacity of 9812 %. At the same time, the advantages and disadvantages of two kinds of building methods are discussed
