从4个月胎肝中制备提取物,采用3HTdR掺入、MTT比色法和细胞形态学等技术观察了胎肝提取物(ELE)对BEL7402肝癌细胞的影响。1%和2%DMSO对细胞DNA合成的抑制率为56%和56%;对线粒体脱氢酶的抑制率为43%和48%;形态学改变主要表现为有丝分裂相减少,胞质伸出粗细不等的突起,相互连成网状结构。50500mg/L的EHE可明显抑制7402细胞的DNA合成,并呈剂量作用效应,IC50为240mg/L;对线粒体脱氢酶活力的影响,500mg/L时有轻微的抑制,而50mg/L则有一定的促进作用;对7402细胞形态改变与DMSO作用后分化的形态改变相似。以上结果提示,ELE可抑制7402细胞的增殖并促进其分化。
