应用UPLC-Q-TOF-MS技术和血清药物化学理论分析检测芩百清肺浓缩丸的入血成分。灌胃给予大鼠芩百清肺浓缩丸溶液后,肝门静脉采血,血清经SPE处理,采用ACQUITY UPLC BEH C_(18)色谱柱(2.1 mm×100 mm,1.7μm),0.1%甲酸水(A)-0.1%甲酸乙腈(B)流动相系统进行梯度洗脱,流速0.3 m L·min~(-1),柱温35℃。通过对比分析体外样品、空白血清及含药血清,同时结合Peakview和Metabolitepilot软件鉴定和表征其血中移行成分,在给药后的大鼠血清中分析鉴定了28个血中移行成分,其中18个原型成分,10个代谢产物。应用UPLC-Q-TOF-MS技术和血清药物化学理论较为全面地阐明了芩百清肺浓缩丸在大鼠血清中的移行成分,为进一步研究其体内代谢过程以及药效物质基础提供了科学依据。
UPLC-Q-TOF-MS technology and serum drug chemistry theory analysis of Qinbai Qingfei Pill into the blood components. The rats were given gavage of Qinbaiqing Pneumoconiosis Pill by intragastric administration. Blood was drawn from the portal vein and the serum was collected by SPE. The samples were separated by ACQUITY UPLC BEH C 18 column (2.1 mm × 100 mm, 1.7 μm), 0.1% formic acid ) - 0.1% formic acid in acetonitrile (B) as mobile phase at a flow rate of 0.3 m L · min ~ (-1) and column temperature of 35 ℃. Through comparative analysis of in vitro samples, blank serum and drug-containing serum, Peakview and Metabolitepilot software were used to identify and characterize the blood migration components, and 28 blood components were identified and analyzed in the rat serum after administration Prototype components, 10 metabolites. The application of UPLC-Q-TOF-MS technology and serum drug chemistry theory comprehensively elucidated the migration components of Qinbai Qingfei Pill in rat serum, which provided a scientific basis for further study of its metabolic process and pharmacodynamic basis .
