利用有丝分裂分子标志———组蛋白H3(Ser10)标记多房棘球蚴成体未分化细胞(neoblast)。同时,利用成体未分化细胞可能的分子标志———miR-71和AGO-2,通过原位杂交方法分析其表达特征。结果显示,多房棘球蚴组蛋白H3阳性细胞没有明显的组织分布特征,而且数量少;AGO-2阳性细胞在组织分布和数量上与组蛋白H3阳性细胞相似,而miR-71阳性细胞在原头蚴中分布广泛,并且在不同发育阶段的原头蚴中miR-71的表达水平存在明显差异。这些研究结果表明,miR-71分子在多房棘球蚴成体未分化细胞中呈非特异表达,不能作为鉴定它的分子标志。
The adult neoplasia of Echinococcus multilocularis was labeled with the mitotic molecular marker Histone H3 (Ser10). Meanwhile, miR-71 and AGO-2, the possible molecular markers of adult undifferentiated cells, were analyzed by in situ hybridization. The results showed that histone H3 positive cells in Echinococcus multilocularis had no obvious histological distribution and a small number of AGO-2 positive cells. The distribution and number of AGO-2 positive cells were similar to that of histone H3 positive cells, whereas miR- There is a wide distribution in the miracidia and there is a significant difference in the expression level of miR-71 in the protoscoleces at different developmental stages. These findings suggest that miR-71 molecules are nonspecifically expressed in adult undifferentiated cells of Echinococcus multilocularis and can not be used as molecular markers to identify them.
