目的分析与N-糖链合成相关β-1,4半乳糖基转移酶-Ⅰ、Ⅱ、Ⅴ(β1,4-galactosyltransferaseⅠ、Ⅱ、Ⅴ,β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ)mRNA在正常脑组织和各级人星形胶质细胞瘤的表达变化。方法采用RT-PCR方法,分析β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在人星形胶质细胞瘤的表达存在。以扩增的3种同工酶cDNA片段作为探针进行标记,Northernblot分析其mRNA在正常脑组织和各级人星形胶质细胞瘤中的表达变化。结果RT-PCR检测到β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在人星形胶质细胞瘤中的表达存在。northernblot发现β-1,4-GalT-Ⅱ、Ⅴ在正常脑组织中有表达,但β-1,4-GalT-Ⅱ的表达较低,而β-1,4-GalT-Ⅰ在正常脑组织中表达极低。β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在星形胶质细胞瘤中的表达显著高于正常脑组织。其中,β-1,4-GalT-Ⅰ、Ⅱ在各级胶质瘤中的表达没有明显差异。而β-1,4-GalT-Ⅴ的表达随胶质瘤分级的增高而增加。结论N-糖链合成相关的β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在人脑星形胶质细胞瘤发生过程中的表达不同,尤其是β-1,4-GalT-Ⅴ的表达与人星型胶质瘤的恶性程度显著相关。
Objective To analyze the expression of β-1,4-galactosyltransferase-Ⅰ, Ⅱ, Ⅴ (β1,4-galactosyltransferaseⅠ, Ⅱ, Ⅴ, β-1,4-GalT-Ⅰ, Ⅱ, Ⅴ) mRNA expression changes in normal brain tissue and human astroglioma at all levels. Methods RT-PCR was used to analyze the expression of β-1,4-GalT-Ⅰ, Ⅱ and Ⅴ in human astrocytoma. The amplified 3 isozyme cDNA fragments were used as probes to detect the expression of mRNA in normal brain tissue and human astroglioma at different levels. Results The expression of β-1,4-GalT-Ⅰ, Ⅱ and Ⅴ in human astrocytomas was detected by RT-PCR. Northernblot showed that β-1,4-GalT-Ⅱ and Ⅴ were expressed in normal brain tissue, but the expression of β-1,4-GalT-Ⅱ was lower than that in normal brain tissue Very low expression. The expression of β-1,4-GalT-Ⅰ, Ⅱ and Ⅴ in astrocytoma was significantly higher than that in normal brain tissue. Among them, β-1,4-GalT-Ⅰ, Ⅱ in all levels of glioma expression no significant difference. The expression of β-1,4-GalT-Ⅴ increased with the increase of glioma grade. Conclusion The expression of β-1,4-GalT-Ⅰ, Ⅱ and Ⅴ related to the N-glycan synthesis in human brain astrocytoma is different, especially the expression of β-1,4-GalT-Ⅴ Significantly correlated with the degree of malignancy of human astrocytomas.
