应用过氧化酶标记的链霉卵白素染色疗法,研究转移抑制基因nm23-H1的表达产物二磷酸核奇激酶(NDPK)在人鼻咽癌中的发达及其意义。结果表明,nm23-H1/NDPK表达与鼻咽癌患者的性别、年龄、临床分期无关。在3例正常鼻咽组织中阳性率为100%;在作癌鼻咽粘膜组织上皮细胞中阳性率为100%(7/7);癌旁组织上皮细胞中阳性率为73.8%(28/38);鼻咽癌组织细胞中,阳性率为44.7%(17/38),差异有显著意义(P<0.05)。另外,nm23-H1/NDPK在鼻咽癌无颈淋巴结转移的病例标本(cN0)中阳性率为75.0%(9/12);在有颈淋巴结转移的病例标本(cN1~3)中阳性率为30.8%(8/26),两者有显著差异(P<0.05)。提示,nm23-H1/NDPK表达与鼻咽癌患者的淋巴结转移与否是显著负相关,可能在鼻咽癌的淋巴结转移过程中起负性调控作用。
Peroxidase-labeled streptavidin staining was used to study the development of NDPK, a metastasis suppressor gene nm23-H1, and its significance in human nasopharyngeal carcinoma. The results showed that, nm23-H1 / NDPK expression and nasopharyngeal carcinoma patients gender, age, clinical stage has nothing to do. The positive rate was 100% in 3 normal nasopharyngeal tissues and 100% (7/7) in epithelial cells of NPC. The positive rate of epithelial cells in paracancerous tissues was 73.8% (28 / 38). In addition, the positive rate of nm23-H1 / NDPK in nasopharyngeal carcinoma without cervical lymph node metastasis (cN0) was 75.0% (9/12), positive in cN1-3 with cervical lymph node metastasis The rate was 30.8% (8/26), with significant difference (P <0.05). These results suggest that there is a significant negative correlation between the expression of nm23-H1 / NDPK and lymph node metastasis in patients with nasopharyngeal carcinoma, which may play a negative regulatory role in the lymph node metastasis of nasopharyngeal carcinoma.
