目的:研究脑康Ⅱ号对糖尿病大鼠学习记忆功能的干预情况。方法:使用脑康Ⅱ号对糖尿病大鼠进行干预,采用水迷宫实验和流式细胞技术,分别观察糖尿病大鼠的行为学改变和海马神经细胞凋亡发生率的变化,并用免疫组化法检测海马CA1区凋亡相关蛋白半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)、B细胞淋巴瘤/白血病-2基因(Bcl-2)的表达水平。结果:脑康Ⅱ号能够显著缩短大鼠Morris水迷宫到达站台时间,并减少大鼠海马神经细胞凋亡率,上调海马CA1区Bcl-2表达,减少Caspase-3表达。结论:脑康Ⅱ号能够通过上调Bcl-2蛋白、下调Caspase-3蛋白的表达,减少海马神经细胞凋亡,改善糖尿病大鼠认知障碍。
Objective: To study the effect of Naokang Ⅱ on learning and memory in diabetic rats. Methods: Nao Kang Ⅱ was used to intervene in diabetic rats. Morris water maze test and flow cytometry were used to observe the changes of behavior and the apoptosis rate of hippocampus neurons in diabetic rats. The expression of Caspase-3, Bcl-2 (Bcl-2) in the hippocampal CA1 region was detected by immunohistochemistry. Result: Naokang Ⅱ could significantly reduce the time to reach the platform of Morris water maze, reduce the apoptosis rate of hippocampal neurons, increase the expression of Bcl-2 and decrease the expression of Caspase-3 in hippocampal CA1 area. Conclusion: Nao Kang Ⅱ can reduce the apoptosis of hippocampal neurons and improve the cognitive impairment of diabetic rats by up-regulating Bcl-2 protein, down-regulating Caspase-3 protein expression.
