几丁质合成酶A作为昆虫蜕皮和变态发育过程中几丁质合成的关键酶,已成为环境友好杀虫剂的理想靶标之一。为研究香梨优斑螟几丁质合成酶的基因结构及潜在应用,以同源基因克隆技术获得目的基因序列,利用生物信息软件进行序列分析。结果显示:该基因5’端2534bp碱基序列编码了蛋白质804个氨基酸,包含催化区域11个保守区中的8个,TMHMM 2.0软件预测此蛋白质中含有9个跨膜螺旋。以13种昆虫氨基酸序列(相似度大于90%)构建系统发育树,结果显示香梨优斑螟(Euzophera.pryiella)与脐橙螟蛾(Amyelois Transitella)进化关系最近,其次云杉卷叶蛾(Choristoneura fumiferana),与三种蝶类(Papilio sp.)及家蚕(Bombyx mori)进化关系较远。
Chitin synthase A, as a key enzyme for chitin synthesis during insect molting and metamorphosis, has become one of the ideal targets for environmentally friendly pesticides. In order to study the gene structure and potential application of chitin synthase in pear, the homologous gene cloning technique was used to obtain the target gene sequence and bioinformatics software was used for sequence analysis. The results showed that the 2534bp base sequence of this gene encoded 804 amino acids of protein, including 8 of 11 conserved regions in the catalytic domain. TMHMM 2.0 software predicted that this protein contained 9 transmembrane helices. Phylogenetic tree was constructed based on the amino acid sequence of 13 insects (similarity greater than 90%). The results showed that Euzophera.pryiella had the closest evolutionary relationship with Amyelois Transitella, followed by Choristoneura fumiferana , And three species of butterfly (Papilio sp.) And Bombyx mori evolutionary relationship.
