目的:探讨中药成分三七总皂苷(PNS)对hERG(human ether-a-go-go related gene)钾离子电流的作用。方法:使用全细胞膜片钳技术观察不同浓度PNS对转染hERG通道的HEK293细胞电流活动的影响。结果:100和300μg·mL-1PNS能够增强转染于HEK293细胞上hERG电流的幅度,并呈时间依赖性。结论:PNS具有增强hERG电流的作用,这种效应可能与PNS影响hERG通道电流的灭活过程有关。
Objective: To investigate the effect of PNS on potassium current in human ether-a-go-go related gene. Methods: Whole-cell patch-clamp technique was used to observe the effect of different concentration of PNS on the activity of HEK293 cells transfected with hERG channel. Results: 100 and 300μg · mL-1PNS could increase the amplitude of hERG current in HEK293 cells in a time-dependent manner. Conclusion: PNS can enhance the hERG current, which may be related to the effect of PNS on the inactivation of hERG channel current.
